My two sons have both. Can you give any more information on who this is for or what you are looking for? A real "shoot from the hip" comparison is the Wii seems more family oriented and the Xbox seems more for Call of Duty and games of that type. We bought the Wii last year for Christmas and it was very popular for both of my boys. As the year went on, my 14 year old became more interested in the Call Of Duty type games. That is a pretty simplistic view, though. If you Google "Wii or Xbox" you can get some more information, too. Some of the posts that it will point you to also mention Wii is more "cartoony" and Xbox is more for serious gamers.
We have the Xbox 360 Kinect. We don't generally use it for FPS games - that's not our style - but rather were using it for physical activity. It's fantastic, especially the freebie games that they included in the unit, Kinect Adventures. My favorite was River Rage.

Note that the only significant criticism in the review was that the games don't have replay value. That's ridiculous. We played the games every other day for several weeks, and never got tired of them (though they are physically demanding, for older folks like us, so I suppose you can say that their replay value is limited by our physical limitations). The games are repetitive just like playing tennis is repetitive, yet they still hold the US Open tournament every year. :shrug:
We're not expert gamers, and really are far more interested in how our gaming system can provide us entertainment (and physical activity), than how our gaming system can provide us challenges - my wife and I both get enough challenge from the work we do. Since we're not inclined to devote ourselves to being hardcore gamers, we find that most of the most popular games are simply too difficult for us. We have played Alan Wake, though, and its stunning on our big screen, and enjoyable, as long as we have the cheat sheets handy.
We have both. The kinect seems to win but the Wii has some things going for it.
The kinect really has no tactile feedback which Wii has. This means if you do an action on the screen you won't feel anything. Wii remote (wiimote) vibrates.
That said, the possibilities with kinect are endless.

I don't have a gaming system but I will admit to being more fascinated by the Wii than any of the others out there. If I were ever to get one, it would probably be that.

Well the Xbox 360 itself has better games than the Wii, but I have not been impressed by the tediousness of the Kinect so far.
We got the Kinect in December. Everyone here loves it. The kids, DH and myself all use it. We love the Your Shape games. They have gym games that everyone loves to play and it is set up for up to 4 players. Great workout while having fun with the kids.
